Prolamins from wheat are named gliadins.

The use of gluten in foodstuffs is extremely common because of its useful effects on e.g. texture, moisture retention and flavor. However, gluten intolerance disorders like coeliac disease require a permanent gluten-free diet to avoid clinical symptoms. According to the Codex Alimentarius (CODEX STAN 118/1979) two categories for labelling of dietary food according to the gluten content exist:

1) "Glutenfree" = Food products, containing < 20 mg / kg gluten
2) "Very low gluten" = Food products containing 20 - 100 mg / kg gluten

The threshold of 20 mg / kg has been adopted by many national legislations in many countries. The Méndez method – a combination of R5 ELISA and a special, patented buffer (cocktail) for optimized extraction of gliadin from heat-processed and non-heated food samples – is the Codex Alimentarius recommended method for analysis of gluten in food. It is Codex Alimentarius type I method for analysis of gliadin/gluten. Categorization as type I method defines it as reference method. R-Biopharm is the only R5 ELISA manufacturer who offers the licensed Cocktail (patented) (Art. No. R7006 / R7016) and hence fulfills the requirements of the reference method to 100 %.

Furthermore, RIDASCREEN® Gliadin is in combination with the Cocktail (patented) the only R5 ELISA, which is AOAC Official Method of Analysis (OMA 2012.01). Applicability for analysis of principally all foods is an important requirement to function as Méndez method in the meaning of Codex Alimentarius. The publication of the 3rd international collaborative proficiency study from 2020 describes the preparation of 900 samples of 19 differently processed foods from 16 food categories and the obtained results demonstrating it suitability for gluten detection in gluten-free foods. Accordingly, RIDASCREEN® Gliadin received from the AOAC as first assay worldwide the status as Official Method of Analysis (AOAC OMA 2012.01) “in food” in 2021. A limitation on certain validated food matrices as usual for other AOAC approvals is not present any longer.

Further approvals:

The RIDASCREEN® Gliadin in combination with the Cocktail (patented) is not only Codex Alimentarius type I method and AOAC Official Method of Analysis. The usage of soya as food and feed has strongly increased in the last decades, leading to an increase in allergic reactions as well.

Benefits:

R7103 is the only kit that detects highly heated soy proteins.

Detects native and processed soy proteins.

Same antibody used as in RIDASCREEN®FAST Soya (R7102).

The dip stick RIDA®QUICK Soya shows no cross reactivity.

No hazardous material is used.

.

Intended use:

RIDASCREEN®FAST Lupine is a sandwich enzyme immunoassay for the quantitative analysis of lupine proteins in food (e.g. juices, wine, beer, sausages, noodles, chocolate, bakery products or ice cream).

Specifications:

Test format: Microtiter plate with 48 wells (6 strips with 8 removable wells each)

Sample preparation: Homogenization, extraction and centrifugation

Incubation time: 30 min, Room temperature

LOD (Detection limit):
0.32 mg / kg (ppm) lupine protein (mean)
0.12 – 0.65 mg / kg (ppm) lupine protein*
*depending on matrix

LOQ (Limit of quantification): 1 mg / kg (ppm) lupine protein

Cross reactivity:
A slight cross reactivity exists to chickpea, soy flour, roasted hazelnut, curcuma, curry and fenugreek.

Validated matrices: Juices, wine, beer, sausages, noodles, chocolate, bakery products or ice cream.

Detected analyte:

Sweet lupine proteins, including the y-conglutin.

All European species of sweet lupines (Lupinus albus, luteus and angustifolius).

Evaluation: Microtiter plate spectrophotometer (450 nm)

General Information:

The macroscopic granular sweet lupines contain 35 - 42 % proteins in the seeds. Lupines are generally used as a natural protein source as an alternative to soy and animal products in human food and animal feed. They are able to bind water in food and therefore can be used as a replacement of egg in bread and bakery products. This increases the freshness and stability of those products. Beside this, lupine flour is used instead of wheat, rye or barley flour in the case of dietetic food. In France it is allowed to mix up to 10 % lupine flour to other flours without further declaration. Due to the fact that lupines can cause severe allergic reactions they should be declared in food products. The allergen can be present as an ingredient or as a contamination in raw and cooked products. According to the regulation (EU) No. 1169/2011 lupine must be declared on food labels.

Benefits:

The test detects all proteins of the European sweet lupine genera used in food, including the heat-stable conglutin y-gamma.

Widespread testing for cross reactivity (89 matrices have been evaluated).

Intended use:

RIDA®QUICK Gliadin (ready to swab) can be used as a swab test for the gluten detection on surfaces in the hygiene control and in cleansing waters (CIP water). So to speak, peanut is one of the most popular “nuts” worldwide. Peanuts are very common ingredients of chocolate products and also consumed as snacks and in bakery products such as cookies. In 2023, 50 million tons peanut were produced worldwide, stating the importance of peanut as a food product.

With 25 %, the fraction of proteins in peanuts is very high. Many of these proteins are known for being allergenic, such as Arachins and Conarachins, which are contained in relative high amounts. Peanut is also one of the five foods, which are the most frequent cause of food allergies. Allergies against peanuts are among the most severe allergies and can lead to life-threatening anaphylactic shocks. Consumption of probably less than a milligram of peanut can induce allergic reactions in very sensitized individuals. Therefore, labeling of peanut containing products is very important and compulsory in the USA and the European Union as well as in many other countries. The main threat for sensitized individuals is contamination of peanut in food, which is intended to be peanut-free (e.g. peanut traces in chocolate, which should not contain peanuts). Until now, there is no cure for food allergies. The only effective treatment is a diet with strict avoidance of foods containing peanuts. Therefore, it is even more important to avoid cross contamination, which can lead to accidental consumption by allergic patients. If an allergen is present as a product ingredient, it must be indicated on the food label according to the regulation (EU) No. 1169/2011. Similar regulations exist e.g. in the USA, Canada, Australia and New Zealand.

To avoid allergic reactions, the safety of foods is ensured by the labeling of allergenic ingredients. Nevertheless, a high risk is still an unintended presence of allergens by cross-contamination due to the food process. It is therefore important to avoid and detect cross-contamination (e.g. due to residues on production lines). The bioavid rapid tests can be used for various applications to detect cross-contamination. For example, incoming raw materials or the effectiveness of the cleaning of production facilities can be checked.

An important challenge with the safe and fast LFD analysis are false negative results due to very high allergen concentrations in the test sample. The so-called “high dose hook effect” (or “overload effect”; Standard EN 15633-1. 2019) is observed when a very high amount of an analyte is present in the sample. In this case, the development of the test line can be inhibited and there is a risk that the test result will be falsely evaluated as negative (or analyte undetectable). The hook line on the test strip makes this effect visible. A missing hook line indicates a high allergen content in the sample and can no longer be evaluated as negative. The evaluation card for the bioavid test strips with hook line can be downloaded here.

The bioavid dip sticks are an easy-to-use method and enable a quick and reliable analysis. Even though most children eventually outgrow their allergy to milk, milk allergy is also among the most common food allergies in adults. Cow’s milk contains several proteins that have an allergic potential. The most important ones are casein and B-lactoglobulin. Cow’s milk contains 3.2 % proteins, which consist of approx. 10 % B-lactoglobulin (leading protein of whey), approx. 80 % caseins and approx. 10 % other proteins.

It is important to note that milk allergy is not the same as lactose intolerance. The cause of lactose intolerance is a deficiency of an enzyme that degrades lactose (milk sugar). Milk allergy, on the other hand, is a true food allergy, which is caused by allergenic constituents of milk proteins. Milk is consumed directly, as a drink, or as a processed product such as cream, butter, yogurt or cheese. Cow’s milk powder is widely used in baked goods, sauces and dessert. Casein can be present as an ingredient or as a contaminant in raw and processed food products. Skim milk powder or caseins/caseinates are often added to food products (e.g. in sausages) to increase the protein content or as thickening agent.Until now, there is no cure for food allergies. The only effective treatment is a diet with strict avoidance of foods containing milk. Therefore, it is even more important to avoid cross contamination, which can lead to accidental consumption by allergic patients. Milk allergy is an allergy in the classic sense, i.e. the immune system fights a substance that is actually harmless to the body and allergic symptoms arise. Milk is consumed directly, as a drink, or as a processed product such as cream, butter, yogurt or cheese. Cow’s milk powder is widely used in baked goods, sauces and dessert.

Cow`s milk contains 3.2 % proteins which consist of 10 % B-lactoglobulin (leading protein of whey) and 80 % caseins. Casein is heat-stable, while B-lactoglobulin is heat-sensitive. Most milk allergy sufferers are allergic to both protein fractions in milk. The most important allergen for children is B-lactoglobulin while the caseins become to be dominant later in adults. The allergen can be present as an ingredient or as a contamination in raw and cooked products. According to the regulation (EU) No. 1169/2011 milk and products thereof must be declared on food labels. Similar regulations exist e.g. in the USA, Canada, Australia and New Zealand.

Milk contains approx. 3.2 % milk protein. Gluten is a mixture of prolamin and glutelin proteins present in wheat, rye and barley. Coeliac disease is a permanent intolerance to gluten that results in damage to the small intestine and is reversible when gluten is avoided by diet. According to the Codex Alimentarius (CODEX STAN 118/1979) two categories for labeling of food according to the gluten content now exist:
1.) “Glutenfree” = Food products, containing < 20 mg / kg Gluten
2.) “Very low gluten” = Food products containing 20 – 100 mg / kg Gluten
The threshold of 20 mg / kg has been adopted by many national legislations in many countries.

During food processing like e.g. fermentation or hydrolysis intact prolamin molecules are partly or completely degraded to small peptide fragments. For Coeliac patients these fragments still remain dangerous even after digestion in the stomach. Single, small peptide sequences (motifs) cannot be detected by a sandwich ELISA format, because at least two epitopes are necessary for a sandwich ELISA. However, single peptide fragments can be detected with a competitive format.

Approvals:

The ELISA RIDASCREEN® Gliadin competitive was evaluated by 18 labs in an international collaborative study (AACCI and WGPAT) for beer, starch syrup and sourdough. Egg white contains approx. 9 - 11 % protein. Four main allergenic proteins come to 80 % of the egg white proteins. These main allergens are ovomucoid (11 %), ovalbumin (54 %), ovotransferrin (12 %) and lysozyme (3.5 %). The allergenic potential of the proteins present in egg yolk is only moderate. Until now, there is no cure for food allergies. The only effective treatment is a diet with strict avoidance of foods containing egg. Therefore, it is even more important to avoid cross contamination, which can lead to accidental consumption by allergic patients. According to the regulation (EU) No. 1169/2011, egg and products thereof must be declared on food labels. Similar regulations exist e.g. in the USA, Canada, Australia, China and New Zealand.

The application is very simple and can also be carried out by personnel without laboratory education. All test kits can be stored at room temperature and are therefore an ideal tool for direct testing in the field, in production facilities or for rapid food testing.

Strongly colored (e.g. coffee, tea, spices, wine), polyphenol / tannin containing, oily or acidic / alkaline samples (e.g. juices) should be extracted with the Absorbent Buffer (Art. Coeliac disease is a permanent intolerance to gluten that results in damage to the small intestine and is reversible when gluten is avoided by diet. According to the Codex Alimentarius (CODEX STAN 118/1979) two categories for labeling of food according to the gluten content now exist:
1.) "glutenfree" = Food products, containing < 20 mg/kg Gluten
2.) "very low gluten = Food products containing 20 - 100 mg/kg Gluten
The threshold of 20 mg/kg has been adopted by many national legislations in many countries.

Traditionally, cereal proteins are subdivided according to their solubility (Osborne fractionation). Unfortunately, the subdivision according to solubility is not an accurate criterion, since co-solubilization and co-precipitation occur frequently. Furthermore, additional factors such as temperature can have a high influence on the result. Especially the subdivision into prolamins and glutelins (which form together the gluten proteins) according to their solubility is very difficult.
Due to the combination of four different monoclonal antibodies, the RIDASCREEN® Total Gluten enables the detection of almost all gluten proteins.
//cdn.shopify.com/s/files/1/0597/5896/9012/files/R7041_a1a3d6ab-3f9c-41de-987d-d7809ae46394.jpg?v=1779863342
surefood®-allergen-shea-nut
SureFood® ALLERGEN Shea Nut

Intended use:

The SureFood® ALLERGEN Shea Nut is a real-time PCR for the direct, qualitative detection of specific shea nut (Vitellaria paradoxa) DNA sequences in raw material.

Specifications:

Test format: 100 reactions

Sample preparation: For DNA preparation the use of SureFood® PREP Advanced (Art. Hazelnuts are very common ingredients of chocolate products and are also consumed as snacks and in bakery products such as cookies. Hazelnut is also one of five foods, which are the most frequent cause of food allergies. In Central Europe, more than 90 % of hazelnut allergies are associated with an allergy to tree pollens. Consumption of a few milligrams of hazelnut can induce allergic reactions in sensitized individuals. Therefore, labelling of hazelnut containing products is very important and compulsory in the USA and the European Union as well as in many other countries. The main threat for sensitized individuals is contamination of hazelnut in food, which is intended to be hazelnutfree (e.g. hazelnut traces in chocolate which should not contain hazelnuts). Therefore, it is important to test foods which may have been contaminated during their production process for the presence of unintended traces of hazelnuts.

Besides being present as an ingredient, the allergen can be present as s a contamination in raw and cooked products. According to the regulation (EU) No. 1169/2011, hazelnut and products thereof must be declared on food labels. Coconut can be bought fresh or as coconut milk, coconut water, creamed coconut or desiccated coconut. It can be used as an ingredient in baking, ice cream, breakfast cereals and confectionary. It is widely used in South-East Asian cooking. Coconut derived products are also added to many food (incl. cakes, chocolates), some infant formulae and to cosmetics and grooming products. Until now, there is no cure for food allergies. The only effective treatment is a diet with strict avoidance of foods containing coconut. Therefore, it is even more important to avoid cross contamination, which can lead to accidental consumption by allergic patients. In 2006, the FDA added coconuts as a "tree nut" to the major food allergens list (Food Allergen Labeling and Consumer Protection Act of 2004 (FALCPA)).

The application is very simple and can also be carried out by personnel without laboratory education. The processing of all bioavid parameters is identical . All test kits can be stored at room temperature and are therefore an ideal tool for direct testing in the field, in production facilities or for rapid food testing.

Strongly colored (e.g. coffee, tea, spices, wine), polyphenol / tannin containing, oily or acidic / alkaline samples (e.g. juices) should be extracted with the Absorbent Buffer. The buffer decolorizes or neutralizes solutions to avoid false positive results and minimize matrix effects.

For the analysis of non-validated samples, the document "SOP Verification Solid Sample Test" (standard operating procedure) can be used. We also offer a paid service for the verification of non-validated matrices.06, the FDA added coconuts as a tree nut to the major food allergens list (Food Allergen Labeling and Consumer Protection Act of 2004 (FALCPA)).

Benefits:

• Better safe than sorry: Included hook line for safe identification of high positive samples
• Uniform procedure for all bioavid parameter
• Easy documentation with RIDA®SMART APP Allergen
• Versatile: applicable for surface-testing, CIP water and food samples (raw and processed)

//cdn.shopify.com/s/files/1/0597/5896/9012/files/BLH700-15_1b455d91-e87d-4011-a59a-7ef7294caa42.jpg?v=1779863471
surefood®-allergen-crustaceans
SureFood® ALLERGEN Crustaceans

Intended use:

The SureFood® ALLERGEN Crustaceans is a real-time PCR for the direct, qualitative and / or quantitative detection of specific crustaceans (Crustacea) DNA sequences according to directive (EC) 1169/2011. They are used as a raw or roasted ingredient in many different foods and can be present as an ingredient or as a contamination. Macadamias belong to the group of tree nuts, which are one of the eight most common allergens. In general, tree nut allergies are common and are an increasingly recognized health concern as availability has increased. Until now, there is no cure for food allergies. The only effective treatment is a diet with strict avoidance of foods containing macadamia. Therefore, it is even more important to avoid cross contamination, which can lead to accidental consumption by allergic patients. According to the regulation (EU) No. 1169/2011, macadamia and products thereof must be declared on food labels. Similar regulations exist e.g. in Canada and Australia.

The application is very simple and can also be carried out by personnel without laboratory education. The processing of all bioavid parameters is identical.

No. BS810-15). The buffer decolorizes or neutralizes solutions to avoid false positive results and minimize matrix effects.

Benefits:

• Better safe than sorry: Included hook line for safe identification of high positive samples
• Uniform procedure for all bioavid parameter
• Easy documentation with RIDA®SMART APP Allergen
• Versatile: applicable for surface-testing, CIP water and food samples (raw and processed)

Intended use:

RIDASCREEN®FAST Soya is a sandwich enzyme immunoassay for the quantitative analysis of native and processed soya proteins in food. The system is specific for roasted peanuts and over detects raw or slightly roasted peanuts.

Specifications:

Test format: Microtiter plate with 96 wells (12 strips with 8 removable wells each)

Sample preparation: Homogenization, extraction and centrifugation

Incubation time: 50 min
Room temperature

LOD (Detection limit): 0.15 mg / kg (ppm) peanut (mean)
0.033 mg / kg (ppm) peanut protein

LOQ (Limit of quantification): 0.75 mg / kg (ppm) peanut
0.166 mg / kg (ppm) peanut protein

Cross reactivity:
No cross reactivity in 91 commodities

Validated matrices: Breakfast cereals, granola bar, cookies, ice cream and chocolate

Detected analyte: Peanut proteins

Evaluation: Microtiter plate spectrophotometer (450 nm)

Related products: SureFood® ALLERGEN Peanut

General Information:

Although peanut (Arachis hypogaea) belongs to the legumes and not to the nuts, its taste and its usual consumption is similar to tree nuts. So to speak, peanut is one of the most popular “nuts” worldwide. Peanuts are very common ingredients of chocolate products and are also consumed as snacks and in bakery products such as cookies.
Therefore, labelling of peanut containing products is very important and compulsory in the USA and the European Union as well as in many other countries. The main threat for sensitized individuals is contamination of peanut in food which is intended to be peanut-free (e.g. peanut traces in chocolate which should not contain peanuts). Therefore, it is important to test foods which may have been contaminated during their production process for the presence of unintended traces of peanuts.
The allergen can be present as an ingredient or as a contamination in raw and cooked products. According to the regulation (EU) No. 1169/2011, peanut and products thereof must be declared on food labels. Even though most children eventually outgrow their allergy to milk, milk allergy is also among the most common food allergies in adults. Cow’s milk contains several proteins that have an allergic potential. The most important ones are casein and ß-lactoglobulin. Cow’s milk contains 3.2 % proteins, which consist of approx. 10 % ß-lactoglobulin (leading protein of whey), approx.

Skim milk powder or caseins/caseinates are often added to food products (e.g. in sausages) to increase the protein content or as thickening agent. Until now, there is no cure for food allergies. The only effective treatment is a diet with strict avoidance of foods containing milk. Therefore, it is even more important to avoid cross contamination, which can lead to accidental consumption by allergic patients. According to the regulation (EU) No. 1169/2011, milk and products thereof must be declared on food labels.

The application is very simple and can also be carried out by personnel without laboratory education. The processing of all bioavid parameters is identical . All test kits can be stored at room temperature and are therefore an ideal tool for direct testing in the field, in production facilities or for rapid food testing.

Strongly colored (e.g. coffee, tea, spices, wine), polyphenol / tannin containing, oily or acidic / alkaline samples (e.g. juices) should be extracted with the Absorbent Buffer (Art. No.

//cdn.shopify.com/s/files/1/0597/5896/9012/collections/R_biopharm_logo_square_2f3d7246-afe1-4492-a339-ae6f0e0e15aa.jpg?v=1756950224
